NM_001453.3(FOXC1):c.782A>G (p.Asp261Gly) was classified as Uncertain significance for Axenfeld-Rieger syndrome type 3 by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This variant has not been reported in the literature in individuals affected with FOXC1-related conditions. This variant is present in population databases (rs751410971, gnomAD 0.04%), including at least one homozygous and/or hemizygous individual. This sequence change replaces aspartic acid, which is acidic and polar, with glycine, which is neutral and non-polar, at codon 261 of the FOXC1 protein (p.Asp261Gly).
