This variant lies in the MYL2 gene (transcript NM_000432.4) at coding-DNA position 166 through 13 bases into the intron immediately after coding-DNA position 169, deleting this region. Submitter rationale: This variant results in the deletion of part of exon 3 (c.166_169+13del) of the MYL2 gene. It is expected to disrupt RNA splicing. Variants that disrupt the donor or acceptor splice site typically lead to a loss of protein function (PMID: 16199547), however the current clinical and genetic evidence is not sufficient to establish whether loss-of-function variants in MYL2 cause disease. This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with MYL2-related conditions.
