Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_206933.4(USH2A):c.11054G>C (p.Trp3685Ser), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tryptophan with serine at codon 3685 of the USH2A protein (p.Trp3685Ser). The tryptophan residue is highly conserved and there is a large physicochemical difference between tryptophan and serine. This variant is present in population databases (rs749154377, ExAC 0.01%). This variant has not been reported in the literature in individuals affected with USH2A-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive.
