Uncertain significance for Catecholaminergic polymorphic ventricular tachycardia 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006073.4(TRDN):c.1624+1G>A, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the TRDN gene (transcript NM_006073.4) at the canonical splice donor site of the intron immediately after coding-DNA position 1624, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects a donor splice site in intron 28 of the TRDN gene. However, it is currently unclear if variants that occur in this region of the gene cause disease. This variant is present in population databases (rs748011906, gnomAD 0.007%). This variant has not been reported in the literature in individuals affected with TRDN-related conditions. ClinVar contains an entry for this variant (Variation ID: 2152664). Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
