Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003042.4(SLC6A1):c.434C>T (p.Ser145Phe), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SLC6A1 gene (transcript NM_003042.4) at coding-DNA position 434, where C is replaced by T; at the protein level this means replaces serine at residue 145 with phenylalanine — a missense variant. Submitter rationale: This sequence change replaces serine, which is neutral and polar, with phenylalanine, which is neutral and non-polar, at codon 145 of the SLC6A1 protein (p.Ser145Phe). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of SLC6A1-related conditions (PMID: 29315614). In at least one individual the variant was observed to be de novo. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt SLC6A1 protein function. For these reasons, this variant has been classified as Pathogenic.
