NM_000719.7(CACNA1C):c.2555A>C (p.Glu852Ala) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E852A variant (also known as c.2555A>C), located in coding exon 19 of the CACNA1C gene, results from an A to C substitution at nucleotide position 2555. The glutamic acid at codon 852 is replaced by alanine, an amino acid with dissimilar properties. This amino acid position is well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
