Uncertain significance for Dystonic disorder — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000113.3(TOR1A):c.767G>C (p.Ser256Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with threonine, which is neutral and polar, at codon 256 of the TOR1A protein (p.Ser256Thr). This variant is present in population databases (no rsID available, gnomAD 0.009%). This variant has not been reported in the literature in individuals affected with TOR1A-related conditions. ClinVar contains an entry for this variant (Variation ID: 2151093). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt TOR1A protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
