Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_017849.4(TMEM127):c.191A>G (p.Gln64Arg), citing Ambry Variant Classification Scheme 2023: The p.Q64R variant (also known as c.191A>G), located in coding exon 1 of the TMEM127 gene, results from an A to G substitution at nucleotide position 191. The glutamine at codon 64 is replaced by arginine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
