Uncertain significance for Joubert syndrome 21 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001382391.1(CSPP1):c.692T>C (p.Ile231Thr), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with CSPP1-related conditions. This variant is present in population databases (rs758349545, gnomAD 0.006%). This sequence change replaces isoleucine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 240 of the CSPP1 protein (p.Ile240Thr). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
