Uncertain significance for Severe early-onset pulmonary alveolar proteinosis due to MARS deficiency; Charcot-Marie-Tooth disease axonal type 2U — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004990.4(MARS1):c.2458G>A (p.Gly820Ser), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 820 of the MARS protein (p.Gly820Ser). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with MARS-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
