Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001369.3(DNAH5):c.9685A>G (p.Lys3229Glu), citing Ambry Variant Classification Scheme 2023: The p.K3229E variant (also known as c.9685A>G), located in coding exon 57 of the DNAH5 gene, results from an A to G substitution at nucleotide position 9685. The lysine at codon 3229 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
