Uncertain significance for Severe combined immunodeficiency due to DNA-PKcs deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006904.7(PRKDC):c.8264A>C (p.Lys2755Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces lysine, which is basic and polar, with threonine, which is neutral and polar, at codon 2755 of the PRKDC protein (p.Lys2755Thr). This variant is present in population databases (rs748466033, gnomAD 0.009%). This variant has not been reported in the literature in individuals affected with PRKDC-related conditions. ClinVar contains an entry for this variant (Variation ID: 2148781). Experimental studies and prediction algorithms are not available or were not evaluated, and the functional significance of this variant is currently unknown. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
