Uncertain significance for Arrhythmogenic cardiomyopathy with wooly hair and keratoderma; Arrhythmogenic right ventricular dysplasia 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004415.4(DSP):c.5697C>G (p.Ile1899Met), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the DSP gene (transcript NM_004415.4) at coding-DNA position 5697, where C is replaced by G; at the protein level this means replaces isoleucine at residue 1899 with methionine — a missense variant. Submitter rationale: This sequence change replaces isoleucine with methionine at codon 1899 of the DSP protein (p.Ile1899Met). The isoleucine residue is highly conserved and there is a small physicochemical difference between isoleucine and methionine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals affected with DSP-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
