Pathogenic variants in this gene have been associated with autosomal recessive pontocerebellar hypoplasia type 1D. This variant introduces a premature termination codon in exon 7 out of 12 and is expected to result in loss of function, which is a known disease mechanism for EXOSC9 in this disorder (PMID: 29727687, 33040083) (PVS1). This variant has been identified in the compound heterozygous state in at least 1 individual reported in the published literature (PMID: 35893425) (PM3) and has a 0.0100% maximum allele frequency in non-founder control populations (https://gnomad.broadinstitute.org/) (PM2). Based on the current evidence, this variant is classified as pathogenic for autosomal recessive pontocerebellar hypoplasia type 1D.
