NM_002471.4(MYH6):c.718G>A (p.Asp240Asn) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D240N variant (also known as c.718G>A), located in coding exon 6 of the MYH6 gene, results from a G to A substitution at nucleotide position 718. The aspartic acid at codon 240 is replaced by asparagine, an amino acid with highly similar properties. This variant was reported in individual(s) with features consistent with MYH6-related congenital heart disease (Sun H et al. Front Genet, 2020 Nov;11:593688). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
