Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001017420.3(ESCO2):c.160C>T (p.His54Tyr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine, which is basic and polar, with tyrosine, which is neutral and polar, at codon 54 of the ESCO2 protein (p.His54Tyr). This variant is present in population databases (rs769799908, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with ESCO2-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
