Uncertain significance for Dilated cardiomyopathy 1G; Autosomal recessive limb-girdle muscular dystrophy type 2J — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001267550.2(TTN):c.4991G>A (p.Arg1664Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces arginine with glutamine at codon 1664 of the TTN protein (p.Arg1664Gln). There is a small physicochemical difference between arginine and glutamine. This variant is present in population databases (rs375029799, ExAC 0.01%). This variant has not been reported in the literature in individuals with TTN-related disease. This variant identified in the TTN gene is located in the Z band of the resulting protein (PMID: 25589632). It is unclear how this variant impacts the function of this protein. Algorithms developed to predict the effect of missense changes on protein structure and function are unavailable for the TTN gene. In summary, this variant is a rare missense change with unknown impact on protein function.
