NM_078470.6(COX15):c.794A>G (p.His265Arg) was classified as Uncertain significance by GeneDx, citing GeneDx Variant Classification (06012015): p.His265Arg (CAT>CGT): c.794 A>G in exon 6 of the COX15 gene (NM_078470.4). The H265R missense change in the COX15 gene has not been published as a mutation, nor has it been reported as a benign polymorphism to our knowledge. The amino acid change is conservative in that both Histidine and Arginine are positively charged amino acid residues and this change occurs at a position that is not highly conserved in the COX15 protein. However, in-silico algorithms are not consistent in their predictions of whether H265R is damaging to the structure/function of the protein. Therefore, based on the currently available information, it is unclear whether H265R is a disease-causing mutation or a rare benign variant. The variant is found in MITONUC-MITOP panel(s).
