NM_000033.4(ABCD1):c.1794G>T (p.Met598Ile) was classified as Uncertain significance by Women's Health and Genetics/Laboratory Corporation of America, LabCorp, citing LabCorp Variant Classification Summary - May 2015: Variant summary: ABCD1 c.1794G>T (p.Met598Ile) results in a conservative amino acid change to a non-conserved residue located in the ABC transporter-like, ATP-binding domain (IPR003439) of the encoded protein sequence. Four of five in-silico tools predict a benign effect of the variant on protein function. The variant allele was found at a frequency of 3.2e-05 in 158353 control chromosomes (gnomAD). The available data on variant occurrences in the general population are insufficient to allow any conclusion about variant significance. c.1794G>T has been reported in the literature in an individual affected with neurological conditions without strong evidence of causality (Mederer_2020). This report does not provide unequivocal conclusions about association of the variant with Adrenoleukodystrophy. To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. One clinical diagnostic laboratory has submitted a clinical-significance assessment for this variant to ClinVar after 2014, and classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
