Uncertain significance for Ehlers-Danlos syndrome, kyphoscoliotic type, 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_017946.4(FKBP14):c.53T>C (p.Ile18Thr), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces isoleucine, which is neutral and non-polar, with threonine, which is neutral and polar, at codon 18 of the FKBP14 protein (p.Ile18Thr). This variant is present in population databases (rs140239991, gnomAD 0.004%). This variant has not been reported in the literature in individuals affected with FKBP14-related conditions. ClinVar contains an entry for this variant (Variation ID: 2141632). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant¬†is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
