Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_138801.3(GALM):c.280G>A (p.Glu94Lys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamic acid, which is acidic and polar, with lysine, which is basic and polar, at codon 94 of the GALM protein (p.Glu94Lys). This variant is present in population databases (rs114353506, gnomAD 0.1%). This variant has not been reported in the literature in individuals affected with GALM-related conditions. ClinVar contains an entry for this variant (Variation ID: 2140428). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt GALM protein function with a negative predictive value of 80%. Experimental studies have shown that this missense change does not substantially affect GALM function (PMID: 30910422).
