This variant lies in the TRPM4 gene (transcript NM_017636.4) at coding-DNA position 430, where C is replaced by T; at the protein level this means replaces arginine at residue 144 with tryptophan — a missense variant. Submitter rationale: The p.R144W variant (also known as c.430C>T), located in coding exon 4 of the TRPM4 gene, results from a C to T substitution at nucleotide position 430. The arginine at codon 144 is replaced by tryptophan, an amino acid with dissimilar properties. This alteration has been reported in a Brugada syndrome cohort (Liu H et al. PLoS One, 2013 Jan;8:e54131). This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis.
