Uncertain significance for Autosomal dominant nocturnal frontal lobe epilepsy 5; Developmental and epileptic encephalopathy, 14 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_020822.3(KCNT1):c.1770-7_1785dup, citing Invitae Variant Classification Sherloc (09022015): This sequence change falls in intron 17 of the KCNT1 gene. It does not directly change the encoded amino acid sequence of the KCNT1 protein. This variant is present in population databases (rs765358427, gnomAD 0.002%). This variant has not been reported in the literature in individuals affected with KCNT1-related conditions. This variant is also known as p.Ile596Cysfs*11. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
