The glutamine amino acid residue is found in multiple mammalian species, which suggests that this missense change does not adversely affect protein function. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This variant disrupts the p.Arg840 amino acid residue in CTC1. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 22267198, 23869908, 24115768, 29228254, 29481669). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. This variant has not been reported in the literature in individuals affected with CTC1-related conditions. This variant is present in population databases (rs776840732, gnomAD 0.006%).
