This variant lies in the MECP2 gene (transcript NM_001110792.2) at coding-DNA position 634, where A is replaced by G; at the protein level this means replaces lysine at residue 212 with glutamic acid — a missense variant. Submitter rationale: In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces lysine, which is basic and polar, with glutamic acid, which is acidic and polar, at codon 200 of the MECP2 protein (p.Lys200Glu). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with MECP2-related conditions (PMID: 26852508). In at least one individual the variant was observed to be de novo. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt MECP2 protein function.
