This variant lies in the TGFBR1 gene (transcript NM_004612.4) at coding-DNA position 696, where G is replaced by C; at the protein level this means replaces lysine at residue 232 with asparagine — a missense variant. Submitter rationale: p.Lys232Asn (AAG>AAC): c.696 G>C in exon 4 of the TGFBR1 gene (NM_004612.2) The Lys232Asn variant in the TGFBR1 gene has not been reported as a disease-causing mutation or as a benign polymorphism to our knowledge. Lys232Asn results in a semi-conservative amino acid substitution of a positively charged Lysine with a neutral, polar Asparagine at a position that is conserved across species. In silico analysis predicts Lys232Asn is probably damaging to the protein structure/function. Mutations in this codon (Lys232Glu) and in nearby residues (Glu218Lys, Phe234Leu) have been reported in association with Loeys-Dietz syndrome (LDS), further supporting the functional importance of this region of the protein. Furthermore, the Lys232Asn variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations.
