This variant lies in the CDKL5 gene (transcript NM_001323289.2) at coding-DNA position 100, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 34 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This premature translational stop signal has been observed in individual(s) with clinical features of CDKL5-related conditions (PMID: 27779742). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Glu34*) in the CDKL5 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in CDKL5 are known to be pathogenic (PMID: 22872100).
