Pathogenic — the classification assigned by GeneDx to NM_181486.4(TBX5):c.421_443delinsCCAGGA (p.Ser141fs), citing GeneDx Variant Classification (06012015): The c.421_443delTCCCCCGCCACCGGGGCGCATTGinsCCAGGA pathogenic variant in the TBX5 gene causes a frameshift starting with codon Serine 141, changes this amino acid to a Proline residue and creates a premature Stop codon at position 36 of the new reading frame, denoted p.S141PfsX36. This pathogenic variant is predicted to cause loss of normal protein function either through protein truncation or nonsense-mediated mRNA decay.
