Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001375.3(DNASE2):c.898G>A (p.Val300Met), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, which is neutral and non-polar, with methionine, which is neutral and non-polar, at codon 300 of the DNASE2 protein (p.Val300Met). This variant is present in population databases (rs35851337, gnomAD 0.003%). This variant has not been reported in the literature in individuals affected with DNASE2-related conditions. This variant is also known as Val284Met. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15"). Experimental studies have shown that this missense change affects DNASE2 function (PMID: 19850016, 23019102, 24242851).
