Uncertain significance — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014495.4(ANGPTL3):c.757G>T (p.Gly253Cys), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glycine, which is neutral and non-polar, with cysteine, which is neutral and slightly polar, at codon 253 of the ANGPTL3 protein (p.Gly253Cys). This variant is present in population databases (rs141967457, gnomAD 0.03%). This variant has not been reported in the literature in individuals affected with ANGPTL3-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). Experimental studies are conflicting or provide insufficient evidence to determine the effect of this variant on ANGPTL3 function (PMID: 28385496, 29713054). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
