Uncertain significance — the classification assigned by GeneDx to NM_005902.4(SMAD3):c.260T>C (p.Ile87Thr), citing GeneDx Variant Classification (06012015): p.Ile87Thr (ATC>ACC): c.260 T>C in exon 2 of the SMAD3 gene (NM_005902.3). A variant of unknown significance has been identified in the SMAD3 gene. The I87T variant has not been published as a mutation, nor has it been reported as a benign polymorphism to our knowledge. The I87T variant was not observed in approximately 6,500 individuals of European and African American ancestry in the NHLBI Exome Sequencing Project, indicating it is not a common benign variant in these populations. The I87T variant is a non-conservative amino acid substitution, which is likely to impact secondary protein structure as these residues differ in polarity, charge, size and/or other properties. Additionally, this substitution occurs at a position that is conserved across species and in silico analysis predicts this variant is probably damaging to the protein structure/function. However, missense mutations in nearby residues have not been reported, indicating this region of the protein may be tolerant of change.
