NM_001042492.3(NF1):c.908T>G (p.Leu303Arg) was classified as Likely pathogenic for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L303R variant (also known as c.908T>G), located in coding exon 9 of the NF1 gene, results from a T to G substitution at nucleotide position 908. The leucine at codon 303 is replaced by arginine, an amino acid with dissimilar properties. This variant was reported in individual(s) with features consistent with neurofibromatosis type 1 (Bausch B et al. J Clin Endocrinol Metab, 2006 Sep;91:3478-81; Cassiman C et al. Clin Genet, 2017 Apr;91:529-535). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). Based on the majority of available evidence to date, this variant is likely to be pathogenic.
