Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_006445.4(PRPF8):c.6942C>A (p.Phe2314Leu), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts the p.Phe2314 amino acid residue in PRPF8. Other variant(s) that disrupt this residue have been observed in individuals with PRPF8-related conditions (Invitae), which suggests that this may be a clinically significant amino acid residue. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site. Experimental studies have shown that this missense change affects PRPF8 function (PMID: 28515276). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) has been performed at Invitae for this missense variant, however the output from this modeling did not meet the statistical confidence thresholds required to predict the impact of this variant on PRPF8 protein function. This missense change has been observed in individuals with retinitis pigmentosa (PMID: 11468273, 31087526; Invitae). This variant is not present in population databases (gnomAD no frequency). This sequence change replaces phenylalanine, which is neutral and non-polar, with leucine, which is neutral and non-polar, at codon 2314 of the PRPF8 protein (p.Phe2314Leu).
