Likely pathogenic for Griscelli syndrome type 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_183235.3(RAB27A):c.428T>C (p.Val143Ala), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces valine, which is neutral and non-polar, with alanine, which is neutral and non-polar, at codon 143 of the RAB27A protein (p.Val143Ala). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with Griscelli syndrome type 2 (PMID: 28936583, 33362801). ClinVar contains an entry for this variant (Variation ID: 2137717). Invitae Evidence Modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) indicates that this missense variant is expected to disrupt RAB27A protein function with a positive predictive value of 95%. Experimental studies have shown that this missense change affects RAB27A function (PMID: 33362801). In summary, the currently available evidence indicates that the variant is pathogenic, but additional data are needed to prove that conclusively. Therefore, this variant has been classified as Likely Pathogenic.
