NM_000021.4(PSEN1):c.392A>G (p.His131Arg) was classified as Uncertain significance for Alzheimer disease 3; Pick disease; Acne inversa, familial, 3; Frontotemporal dementia by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Experimental studies have shown that this missense change affects PSEN1 function (PMID: 27930341, 32087291). This missense change has been observed in individuals with early-onset Alzheimer’s disease (PMID: 23638752, 33188256). This variant is not present in population databases (gnomAD no frequency). This sequence change replaces histidine, which is basic and polar, with arginine, which is basic and polar, at codon 131 of the PSEN1 protein (p.His131Arg).
