This variant lies in the PTS gene (transcript NM_000317.3) at the canonical splice donor site of the intron immediately after coding-DNA position 186, where G is replaced by A; at the protein level this means a change at this position may disrupt normal splicing. Submitter rationale: This sequence change affects a donor splice site in intron 3 of the PTS gene. RNA analysis indicates that disruption of this splice site induces altered splicing and may result in an absent or disrupted protein product. This variant is not present in population databases (gnomAD no frequency). Disruption of this splice site has been observed in individual(s) with biopterin-deficient hyperphenylalaninemia (PMID: 11438997, 23138986). This variant is also known as IVS3+1G>A. ClinVar contains an entry for this variant (Variation ID: 2137247). Studies have shown that disruption of this splice site results in skipping of exon 3 and introduces a premature termination codon (PMID: 11438997).
