Likely pathogenic for Multiple endocrine neoplasia, type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001370259.2(MEN1):c.1231G>C (p.Ala411Pro), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces alanine, which is neutral and non-polar, with proline, which is neutral and non-polar, at codon 411 of the MEN1 protein (p.Ala411Pro). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with MEN1-related conditions (PMID: 12699448). It has also been observed to segregate with disease in related individuals. Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is expected to disrupt MEN1 protein function. Experimental studies have shown that this missense change affects MEN1 function (PMID: 21819486).
