Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_000071.3(CBS):c.1238G>A (p.Arg413His), citing Ambry Variant Classification Scheme 2023: The p.R413H variant (also known as c.1238G>A), located in coding exon 12 of the CBS gene, results from a G to A substitution at nucleotide position 1238. The arginine at codon 413 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
