Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000375.3(UROS):c.634T>C (p.Ser212Pro), citing Invitae Variant Classification Sherloc (09022015): This missense change has been observed in individual(s) with congenital erythropoietic porphyria (PMID: 8655129, 22816431). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. Experimental studies have shown that this missense change affects UROS function (PMID: 8655129, 19099412). For these reasons, this variant has been classified as Pathogenic. This variant is present in population databases (rs139388833, gnomAD 0.0009%). This sequence change replaces serine, which is neutral and polar, with proline, which is neutral and non-polar, at codon 212 of the UROS protein (p.Ser212Pro).
