Pathogenic — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_033100.4(CDHR1):c.2087_2090del (p.Asp696fs), citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. This variant disrupts a region of the CDHR1 protein in which other variant(s) (p.Gly703Valfs*138) have been determined to be pathogenic (PMID: 32141364; Invitae). This suggests that this is a clinically significant region of the protein, and that variants that disrupt it are likely to be disease-causing. This premature translational stop signal has been observed in individual(s) with retinitis pigmentosa (PMID: 26261414). This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Asp696Alafs*3) in the CDHR1 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 164 amino acid(s) of the CDHR1 protein.
