NM_004560.4(ROR2):c.2278C>T (p.Gln760Ter) was classified as Pathogenic by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015). This variant lies in the ROR2 gene (transcript NM_004560.4) at coding-DNA position 2278, where C is replaced by T; at the protein level this means converts the codon for glutamine at residue 760 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This premature translational stop signal has been observed in individual(s) with autosomal dominant brachydactyly type B (PMID: 10986040, 23238279). In at least one individual the variant was observed to be de novo. It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Gln760*) in the ROR2 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 184 amino acid(s) of the ROR2 protein.
