Submitter rationale: For these reasons, this variant has been classified as Pathogenic. Experimental studies have shown that this missense change affects HR function (PMID: 17609203, 26680117). Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. This missense change has been observed in individual(s) with autosomal recessive HR-related conditions (PMID: 10772391; Invitae). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. This variant is present in population databases (rs778999706, gnomAD 0.008%). This sequence change replaces cysteine, which is neutral and slightly polar, with glycine, which is neutral and non-polar, at codon 622 of the HR protein (p.Cys622Gly).
