Pathogenic for Long QT syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000238.4(KCNH2):c.529G>T (p.Glu177Ter), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the KCNH2 gene (transcript NM_000238.4) at coding-DNA position 529, where G is replaced by T; at the protein level this means converts the codon for glutamic acid at residue 177 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: For these reasons, this variant has been classified as Pathogenic. This premature translational stop signal has been observed in individual(s) with long QT syndrome (PMID: 23351921). This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Glu177*) in the KCNH2 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in KCNH2 are known to be pathogenic (PMID: 10973849, 19862833).
