This premature translational stop signal has been observed in individual(s) with 3-Methylglutaconic aciduria, dystonia–deafness, hepatopathy, encephalopathy, Leigh-like syndrome (MEGDHEL) syndrome (PMID: 29205472). For these reasons, this variant has been classified as Pathogenic. This sequence change creates a premature translational stop signal (p.Lys104*) in the SERAC1 gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in SERAC1 are known to be pathogenic (PMID: 22683713).
