This variant lies in the TULP1 gene (transcript NM_003322.6) at 5 bases into the intron immediately after coding-DNA position 999, where G is replaced by C. Submitter rationale: This variant has been observed in individual(s) with TULP1-related conditions (PMID: 18055821, 29178642). In at least one individual the data is consistent with being in trans (on the opposite chromosome) from a pathogenic variant. It has also been observed to segregate with disease in related individuals. This variant is not present in population databases (gnomAD no frequency). This sequence change falls in intron 10 of the TULP1 gene. It does not directly change the encoded amino acid sequence of the TULP1 protein. It affects a nucleotide within the consensus splice site. Variants that disrupt the consensus splice site are a relatively common cause of aberrant splicing (PMID: 17576681, 9536098). Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may disrupt the consensus splice site. For these reasons, this variant has been classified as Pathogenic.
