Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.811A>C (p.Thr271Pro), citing Ambry Variant Classification Scheme 2023: The p.T271P variant (also known as c.811A>C), located in coding exon 6 of the FBN2 gene, results from an A to C substitution at nucleotide position 811. The threonine at codon 271 is replaced by proline, an amino acid with highly similar properties, and is located in the cb EGF-like #01 domain. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
