Uncertain significance for Joubert syndrome; Meckel-Gruber syndrome — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_015272.5(RPGRIP1L):c.1040G>A (p.Arg347Lys), citing Invitae Variant Classification Sherloc (09022015): Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This sequence change replaces arginine, which is basic and polar, with lysine, which is basic and polar, at codon 347 of the RPGRIP1L protein (p.Arg347Lys). This variant is not present in population databases (gnomAD no frequency). This variant has not been reported in the literature in individuals affected with RPGRIP1L-related conditions. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
