Uncertain significance for Neuropathy, hereditary sensory and autonomic, type 2A; Generalized epilepsy with febrile seizures plus, type 7 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_001365536.1(SCN9A):c.2308G>C (p.Glu770Gln), citing Invitae Variant Classification Sherloc (09022015). This variant lies in the SCN9A gene (transcript NM_001365536.1) at coding-DNA position 2308, where G is replaced by C; at the protein level this means replaces glutamic acid at residue 770 with glutamine — a missense variant. Submitter rationale: This sequence change replaces glutamic acid, which is acidic and polar, with glutamine, which is neutral and polar, at codon 759 of the SCN9A protein (p.Glu759Gln). In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated. This variant has not been reported in the literature in individuals affected with SCN9A-related conditions. This variant is not present in population databases (gnomAD no frequency).
