Submitter rationale: This sequence change replaces glycine, which is neutral and non-polar, with serine, which is neutral and polar, at codon 740 of the ELN protein (p.Gly740Ser). This variant also falls at the last nucleotide of exon 31, which is part of the consensus splice site for this exon. This variant is present in population databases (rs375579231, gnomAD 0.02%). This missense change has been observed in individual(s) with tetralogy of Fallot (PMID: 39402625). ClinVar contains an entry for this variant (Variation ID: 213183).
