Pathogenic for Arrhythmogenic cardiomyopathy with wooly hair and keratoderma; Arrhythmogenic right ventricular dysplasia 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_004415.4(DSP):c.1789T>C (p.Ser597Pro), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces serine, which is neutral and polar, with proline, which is neutral and non-polar, at codon 597 of the DSP protein (p.Ser597Pro). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with clinical features of autosomal dominant DSP-related conditions (Invitae). In at least one individual the variant was observed to be de novo. ClinVar contains an entry for this variant (Variation ID: 2131612). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. This variant disrupts the p.Ser597 amino acid residue in DSP. Other variant(s) that disrupt this residue have been determined to be pathogenic (PMID: 20940358; Invitae). This suggests that this residue is clinically significant, and that variants that disrupt this residue are likely to be disease-causing. For these reasons, this variant has been classified as Pathogenic.
